چکیده
Introduction: Catecholamines are charged neurotransmitters found in the neuroendocrine cells called chromaffins. The release of catecholamine, through a narrow fusion pore of a chromaffin cell requires a charge compensation by other ions to maintain an osmotic balance of charges. The aim of this research is to show that the exocytotic catecholamine release is not associated with the cation flux through the channels in the vesicle membrane, but the Na influx (which is present outside the cell) through the fusion pore.

Secretory vesicles membrane area is regulated in tandem with quantal size in chromaffin cells.
The number of transmitter molecules released in a quantal event can be regulated, and recent studies suggest that the modulation of quantal size is associated with corresponding changes in vesicle volume (Colliver et al., 2000; Pothos et al., 2002). If so, this could occur either by distension of the vesicle membrane or by incorporation and removal of vesicle membrane. We performed simultaneous...
متن کاملMyosin II contributes to fusion pore expansion during exocytosis.
During exocytosis, the fusion pore expands to allow release of neurotransmitters and hormones to the extracellular space. To understand the process of synaptic transmission, it is of outstanding importance to know the properties of the fusion pore and how these properties affect the release process. Many proteins have been implicated in vesicle fusion; however, there is little evidence for prot...
متن کاملHeterologous expression of a P2x-purinoceptor in rat chromaffin cells detects vesicular ATP release.
A cloned P2x-purinoceptor was transiently expressed in single isolated rat adrenal chromaffin cells and evaluated for the detection of released ATP. After cytoplasmic injection of the P2x complementary RNA (cRNA; 4-24 h), application of ATP produced an inwardly rectifying current over the voltage range -130 to -10 mV as measured by the whole cell patch-clamp technique. The dose-response curve f...
متن کاملRelationship between fusion pore opening and release during mast cell exocytosis studied with patch amperometry.
We have studied exocytosis in rat peritoneal mast cells by cell-attached patch amperometry. Step increases in capacitance were accompanied by typical amperometric spikes due to the release of 5-hydroxytryptamine (serotonin), indicating exocytosis of typical mast cell granules. We have measured the time course of fusion pore expansion, and correlated it with release from the granule matrix. The ...
